Insulin glargine is an insulin analogue. It is produced by substituting one glycine residue and adding two arginine residues on the double chains of human insulin, hence the name insulin glargine. These structural modifications render the drug solution clear under acidic conditions. When administered subcutaneously into the human body, the near‑neutral physiological environment causes insulin glargine to form fine precipitates, resulting in slow drug release. This mimics the physiological secretion of endogenous insulin, providing stable and effective glycemic control for 24 hours.
Insulin glargine is a newer long‑acting insulin analogue, also referred to as basal insulin. It is injected once daily at a fixed time and exerts a stable glucose‑lowering effect on both fasting and postprandial blood glucose.
